Self-reconstructing Bessel beam created by two-photon-polymerized micro-axicon for light-sheet fluorescence microscopy

Observing micro-organisms with depth-resolving capability is important in optical microscopy for biomedical sciences and industries. We demonstrate the fabrication use of a two-photon polymerized micro-axicon lens that generates self-reconstructing pencil-like Bessel beam light-sheet fluorescence (LSFM), providing 3D internal structures micro-organisms. The fabricated SU-8 100 µm diameter transforms input Gaussian from single-mode fiber into non-diffractive Bessel-Gauss beam. focused spot size 2.3 ± 0.25 long propagation distance over 160 µm, which well-suited LSFM. self-reconstruction generated investigated thoroughly through both simulations experiments. Since this can be directly 3D-printed on fibers’ end facets or small mobile substrates, replace bulky objective conventional microscopes. This will facilitate wide-spread tomographic imaging micro-organisms, especially compact micro-fluidic devices lab-on-a-chip architectures.
